Molecular Detection of (I1307k) Mutation in the Adenomatous Polyposis Coli Among Sudanese Colorectal Cancer Patients, Sudan

Abstract
Background
Several studies have reported that early detection of Adenomatous Polyposis Coli (APC) mutation can improve outcomes and our understanding of the unique biology of colorectal cancer (CRC). This study aimed to identify the (I1307k) Mutation in APC among patients with CRC in Sudan.
Methods
This prospective comparative, laboratory-based study included, archival paraffin-embedded samples obtained from 50 with colorectal cancer and 30 controls.

Molecular assessment was performed. The amplification of (I1307K) APC gene with a length of 79pb was performed using published primers, APC-1, APC1307W, and APC1307M to amplify the gene. Preparation of reaction mixtures, including wild-type and mutant-type mixture amplifications, and visualization of PCR products according to known method in literature.
Results
The study examined 80 samples. Degree of differentiation among cases groups was studied. A significant difference was observed between the APC gene and cancer stage (p=0.022).
The age groups of the study were 23 (28.7%) aged from 20 to 40 years, 31 (38.7%) aged from 41 to 60 years, and 26 (32.5%) were aged > 61 years old with mean (51.5500), with no statistical significance (p=0.937) between APC gene and age. The participants were comprised of 42 (52.5%) males, and 38 (47.5%) females. The association between APC gene and sex showed statistical not significance (p=0.86). The association between APC gen and cancer stage showed statistical significance (p=0.022).
Conclusion
The results of this study showed that there is an association between colorectal cancer and (I1307) mutation in APC. Well differentiated is the predominant abnormality in this study. In addition, no relationship was found between age and APC gene mutation.
